I've had some people respond to me, and some people ignore my responses to their ads.
I recently interviewed with a company for a position completely different than what I responded to, so some companies will take a hard look at what you send them, and see how and where your skills may apply.
At the same time, a couple of months ago, I responded about a posting that I thought I was very qualified for. Years of experience doing the described job, and very positive work experience in the system/city where the project was taking place. Never got a sniff.
